Details
Sirolimus 1mg Tablets is an immunosuppressive agents belonging to the proliferation signal
Inhibitors group.
It is widely used in organ transplantation because of its good response.
It is used as single drug therapy or in combination with other immunosuppressants.
It is used both as prophylaxis and treatment of the organ transplant rejection.
Mechanism of action: It reduces the immunity of the body by binding the circulating immunophilin FK506-binding protein 12 and blocks the formation of mTOR. This mTOR is responsible for the inhibition of T cell proliferation. Sirolimus also inhibits the proliferation of B-cell proliferation and immunoglobulin production.
It is available as Rapamune in both International and Indian markets.
Uses
- Sirolimus 1mg Tablets is used as prophylaxis and treatment of the steroid-refractory acute and chronic GVH reaction in bone marrow transplant recipients.
- It is widely used to prevent kidney transplant rejection.
- It is used with cyclosporine in uveoretinitis and topically in skin infections.
- Rapamune is also used in sporadic lymphangioleiomyomatosis (S-LAM), a rare progressing lung disease.
- It is also used as a coronary stents eluting drug to reduce the restenosis and additional adverse cardiac complications in coronary artery disease patients.
Dosage
- Sirolimus 1mg Tablets come as white-coloured, triangular-shaped, coated tablets in blister packs of 30 and 100 tablets.
- Rapamune must be stored at temperature not exceeding 250C and must be protected from light.
- Final dosages are determined by the doctor on the basis of factors such as medical condition, age, weight and ongoing medications.
- Dose is adjusted to minimise dose related side effects.

- Myelosuppression (especially thrombocytopenia),
- Hepatotoxicity
- Stomach pain, diarrhoea, constipation
- Hypertriglyceridemia
- Pneumonitis
- Hemolytic-uremic syndrome when given in combination with tacrolimus
- Nausea and vomiting
- Increased risk of infections
- Increased blood pressure
- Kidney dysfunction
- Acne
- Joint pain
- Menstrual problems
- Elevated liver function test
Precautions
- Sirolimus 1mg Tablets is contraindicated in case of allergy to sirolimus.
- It must be avoided in pregnancy and breastfeeding.
- Rapamune must be used carefully in case of liver & kidney dysfunction, acute infections, body mass index (BMI) greater than 30 kg/m2 and high risk of kidney rejection.
- Exposure to sunlight must be avoided to reduce the risk of skin cancer.
- Tell the doctor about ongoing medications like immunosuppressants,medicines used to treat infection,high blood pressure medicine,anti-epileptic medicines & medicines for gastrointestinal disorders and any allergy before starting the drug.
Always tell the doctor about pregnancy or future plan of pregnancy or breastfeeding.
