Oxaliplatin 50mg, 100mg Injection is one of the platinum analogs that belongs to the cell cycle non specific (CCNS) anticancer drugs.

It is an inorganic complex having cytotoxic effects and kills cancer cells in all stages of cell cycle.

It is a third-generation diaminocyclohexane platinum analog.

It is effective in a wide range of solid tumors.

Mechanism of action:it acts by getting hydrolysed inside the cell leading to intrastrand and interstrand DNA cross-linking.This results in death of the cancer cell by inhibition of DNA synthesis and function of cancer cell. It also affects nuclear and cytoplasmic proteins which increases the cytotoxicity of this drug.

It is available as Eloxatin in International and as Oxamil in Indian markets.

Uses

  • Oxaliplatin 50mg, 100mg Injection is now approved as first-line treatment of metastatic colorectal cancer along with fluoropyrimidine 5-fluorouracil (5-FU) and leucovorin ( as FOLFOX regimen and with capecitabine as XELOX regimen.
  • Eloxatin is also used in the FOLFOX regimen as adjuvant therapy of stage III and high-risk stage II colon cancer.
  • It is also indicated in other gastrointestinal cancers like gastroesophageal cancer, pancreatic cancer and hepatocellular cancer.
  • Oxamil is widely used in the FOLFIRINOX regimen (intravenous 5-FU, irinotecan, and oxaliplatin) in metastatic pancreatic cancer.

 

Dosage

  • Oxaliplatin 50mg, 100mg Injection comes as a sterile, clear , cloorless, single use vial and must be administered by the healthcare professional only.
  • Oxamil is administered intravenously and pre-hydration is not required like in case of cisplatin.
  • Final dosages are determined by the doctor on the basis of factors such as medical condition, age, weight and ongoing medications.
  • Dose and rate of infusion is adjusted to minimise related side effects.

Precautions

  • Complete blood count, liver function tests and neurologic functions must be monitored regularly.
  • Proper hypersensitivity treatment set up is required as severe hypersensitivity reaction can occur after infusion.
  • Oxaliplatin 50mg, 100mg Injection is avoided in severe myelosuppression, liver dysfunction, congenital long QT syndrome and lung diseases.
  • Tell the doctor about ongoing medications and any allergy before starting the drug.
  • Always tell the doctor about pregnancy or future plan of pregnancy or breastfeeding.
  • It interacts with warfarin.

Oxamil causes severe damage to the foetus in pregnant women. It is contraindicated in pregnant and breastfeeding women.
